First of all, if you need to FLY to the conference, you have a few options.
First, Lancaster DOES have an airport but service may or may not be available. Small municipal airports are struggling to keep commercial carriers interested. If you're interested in checking, the airport (LNS) is commercially covered by "Cape Air" through BWI in Baltimore, MD. The Lancaster Airport website is www.lancasterairport.com.
The second option is Harrisburg International Airport (MDT). Covered by many major airlines, the airport is approximately a 40 minute drive from the hotel via 4 lane, limited access highway (PA283 & US30). All normal services, including car rental, are available.
Your next option might be Philadelphia International Airport (PHL). Again, with most major airlines available and all services. Rand McNally (online) indicates that Philly is 63 miles from the hotel and will be approximately a 1 hour and 40 minute drive.
Finally, from my point of view, is the Baltimore-Washington International Airport (BWI). Reasonably easy access to major highways via the Baltimore Beltway (I495), I83 North (through York, PA) and US30 to the hotel. Again, from Rand McNally, your distance is just under 100 miles and estimated at 1 hour and 50 minutes.

If you're driving, and have a GPS device that allows input of Latitude-Longitude data, the coordinates of The Lancaster Host Resort are
40°01'34.74"N Latitude and 76°12'49.10" W Longitude
The one I use doesn't allow latitude-longitude input but allows me to input the name "Lancaster Host Resort" as a "Point of Interest" and it has found it without problems and the directions were accurate.
